Complete Guide to NAD+ for Metabolic Health
NAD+ (nicotinamide adenine dinucleotide) is a coenzyme involved in cellular energy metabolism. Human studies have examined age-related differences in NAD+ in specific tissues. Dietary supplements containing precursors like NMN or NR are marketed to support normal cellular energy metabolism. NAD+ supplements are not weight loss products.
This article is for educational purposes only and is not medical advice. NAD+ supplements discussed here are dietary supplements, not FDA-approved drugs. IV NAD+ is an unapproved injectable drug, not a supplement, and Boca does not offer it. Talk with your own clinician before starting any supplement.
What Is NAD+?
NAD+ is found in every cell, essential for:
- Energy production (mitochondrial function)
- Metabolic processes (breaking down fats, carbs, protein)
- Supporting normal cellular processes (a role in sirtuin and DNA-repair pathways)
- Cellular health across the lifespan
NAD+ and Age
A 2012 study of human skin tissue found lower NAD+ concentrations associated with older age. It examined samples from 49 surgical patients, including newborns and older participants. This was a comparison between people, not a study tracking each person over time or testing a supplement. It does not establish a universal rate of decline throughout the body, diagnose an individual deficiency or show that supplementation prevents aging.
How to Increase NAD+
Supplements
- NMN (nicotinamide mononucleotide)
- NR (nicotinamide riboside)
Available doses and formats vary by product. Your clinician can help you decide whether a supplement is appropriate for you and what amount makes sense for your situation.

Lifestyle Strategies (Free)
A 2019 human exercise study examined an enzyme called NAMPT, which participates in recycling NAD+. Among 43 younger and older adults who trained for 12 weeks, muscle NAMPT increased after aerobic or resistance training. The measured outcome was enzyme abundance, not proof of a whole-body NAD+ increase or longer life. A separate six-week HIIT cohort did not show changes in the measured NAD+-recycling enzymes in abdominal fat tissue.
This does not establish that HIIT is the best way to raise NAD+, or support a blanket recommendation for fasting, sauna or cold exposure for that purpose. Discuss an appropriate activity plan with your clinician; this article does not prescribe an NAD+-targeted lifestyle regimen.
NAD+ and Weight Loss
NAD+ is a coenzyme involved in metabolism; it is not a weight loss product and should not be expected to cause weight loss.
NAD+ Alongside GLP-1 Medications
Some people who take a prescription GLP-1 medication also ask about NAD+ supplements for general cellular-energy support. These are different categories: a GLP-1 medication is a prescription drug, and NAD+ supplements are dietary supplements.
We do not claim that NAD+ improves how well a GLP-1 medication works. Whether to combine a supplement with any prescription medication is a medical decision. Because supplements can interact with medications, ask the clinician who manages your prescription before adding NAD+ or any supplement.
Who Should Take NAD+?
Age alone does not establish that an NAD+ precursor supplement is appropriate. Discuss your goals, medical conditions and current medicines with your clinician.
A 2021 NIDDK research summary describes a randomized, 10-week trial in 25 postmenopausal women with prediabetes and overweight or obesity. NMN improved muscle insulin sensitivity, but fasting glucose and liver insulin response did not differ significantly between groups. The summary identifies unresolved questions about durability, safety and other populations. This study does not establish a general recommendation for everyone over 40, demonstrate longer human life, or test NR or injectable NAD+.
Safety
NCCIH advises that supplements can cause side effects and interact with medicines. Safety depends on the actual product and your circumstances; this article does not establish the safety of a particular NAD+, NMN or NR preparation. Discuss the product with your clinician, especially during pregnancy or breastfeeding or if you have medical conditions or take medicines. If side effects occur, stop the supplement and contact your healthcare provider. This general guidance does not establish a specific interaction or side-effect rate for NAD+ precursors.
